Is it possible to change my relay address and not lose connectivity with my currently installed Unattended clients?

As long as you have a RelayAddressableUri defined in your web.config, you can make changes to your Relay address as outlined below. This applies to version 4.3 and higher.

Please note, these steps must be done in the correct order!

In this example we are changing both the host name and the port.

1) Make sure the key "RelayAddressableUri" exists in the appSettings section of the web.config, if it does not exist add it and save your changes. The line will look similar to this:
<add key="RelayAddressableUri" value="relay://mydnsname:8041/" />

2) Change the "RelayAddressableUri" to your desired hostname and port and Save your changes. On the host page your Unattended clients will disconnect and then reconnect. Make sure that all Unattended machines have reconnected before continuing to step 3.
<add key="RelayAddressableUri" value="relay://remote.myscreenconnectsite.com:443/" />

3) Once the Unattended clients reconnect to the server select them and then More->Reinstall from the host page. Note that the clients will now appear to be disconnected if you are also changing the port.

4) If needed, modify the key "RelayListenUri" to the port number that matches step 2 and Save your changes.

It's worth noting that there are actually two <appSettings> locations. It will only work when RelayAddressableUri is placed in the same <appSettings> as "WebServerListenUri".
