This is for a self hosted/on prem. install...

I'd rather keep the screen connect website on port 80, but I also have IIS running on the same box, all sites are using host headers and sharing the same IP. Can I still have SC running on the same IP and port 80 too?

For example: <add key="WebServerListenUri" value="http://support.mycompany.com:80/" />

If not, so let's say I have to change the WebServerListenUri to either a unique IP, or a different port, let's say 8080, then do I need to reinstall all access clients? or do they not use/ignore the website on rely only on the RelayListenUri setting?

the easiest way is through piggybacking, which also means one less SSL certificate you need to worry about:
